负载凋亡肿瘤细胞的树突状细胞对喉癌细胞的杀伤作用

Killing effect of apoptotic-tumor-cell-loaded dendritic cells on laryngeal carcinoma cells

摘要: 目的 探讨凋亡肿瘤细胞抗原负载的树突状细胞(DC)对喉癌细胞杀伤作用的影响。方法 用细胞因子重组人白细胞介素4(rhIL-4)、重组人粒细胞巨噬细胞集落刺激因子(rhGM-CSF)与肿瘤坏死因子α(rhTNF-α)体外诱导扩增人外周血单个核来源的DC,6 d后将DC分为凋亡组、冻融组和未处理组;各组细胞继续培养4 h后,分别采用流式细胞术检测DC表面分子的表达、ELISA法检测干扰素γ(IFN-γ)的水平、MTT法评估各组DC刺激细胞毒性T淋巴细胞(CTL)增殖的能力及诱导CTL的杀伤活性。结果 与其余各组相比,凋亡组DC表面分子与IFN-γ的表达明显增加(P<0.05),CTL的增殖能力与杀伤活性增强(P<0.05)。结论 凋亡肿瘤细胞可以促进DC的分化与成熟,并能使DC产生高效而特异性的抗喉癌免疫应答。

Abstract: Objective To explore the killing effect of apoptotic-tumor-cell-loaded dendritic cells (DCs) on the laryngeal carcinoma cells. Methods DCs from peripheral blood monocytes were induced by recombinant human interleukin-4 (rhIL-4), recombinant human granulocyte/macrophage colony-stimulating factor (rhGM-CSF) and tumor necrosis factor (TNF-α) in vitro, then were divided into the apoptotic laryngeal carcinoma cells group, freeze-thaw cells group and untreated cells group 6 days later. After continuous culture of DCs for 4h, the expressions of the surface markers of DCs were detected by flow cytometry, the level of Interferon-γ (IFN-γ) was measured by ELISA, and the proliferation ability and killing effect of cytotoxic T lymphocyte (CTL) stimulated by DCs were evaluated by MTT. Results Compared with other groups, apoptotic laryngeal carcinoma cells group could enhance the expression of the surface markers CD83, CD86, HLA-DR and IFN-γ(P<0.05), and the proliferation ability and killing effect of CTL increased (P<0.05). Conclusion Apoptotic carcinoma cells can promote the differentiation and maturation of DCs, and induce effective and specific immune response against laryngeal carcinoma.
