Expression of DACH1, fatty acid synthase and sterol regulatory element binding protein-1 in endometrial carcinoma

Abstract:

Objective   To study the expression of dachshund (DACH1) protein, fatty acid synthase(FASN) and sterol regulatory element binding protein-1(SREBP1) in endometrial carcinoma and the relation with clinical pathological parameters. Methods   The immunohistochemistry SP method was used to examine the expression of DACH1, FASN and SREBP1 in endometrial carcinoma, endometrial atypical hyperplasias,complex endometrium and normal endometrium. Resluts   The positive rate of DACH1 in endometrial carcinoma was significantly less than that in benign groups, and the positive rates of FASN and SREBP1 in endometrial carcinoma were significantly higher than those in benign groups. The positive rates of DACH1, FASN, SREBP1 in endometrial carcinoma were 40.0%,61.7%,65.0%, and the DACH1, FASN, SREBP1 expressions were all significantly correlated with the differentiated degree.The DACH1 expression was significantly correlated with FIGO staging and histological types of endometrial carcinoma samples, and the SREBP1 expression was also significantly correlated with ages of the endometrial cancer patients. The DACH1 expression was negatively correlated with that of FASN and SREBP1, and the FASN expression was positively correlated with the SREBP1 expression. Single factor analysis showed that patients with positive DACH1 protein expression had a significant better overall survival time compared with patients with negative DACH1 protein expression(P<0.05). Single factor analysis showed that patients with positive FASN and SREBP1 protein expression had a significant poorer overall survival time compared with patients with negative FASN and SREBP1 protein expressions (P<0.05). Conclusion   DACH1, FASN and SREBP1 might be new markers for prognosis prediction of endometrial carcinoma.
