Effects of potassium iodate and bisphenol A on the proliferation of  thyroid carcinoma cells

Abstract:

Objective   To observe the effects of potassium iodate and bisphenol A(BPA)on proliferation of thyroid carcinoma cells BHP10-3. Methods   10-3~10-10mol/L potassium iodate,  10-3~10-10mol/L bisphenol A and 10-7mol/L potassium iodate +10-8mol/L bisphenol A were added to the culture of BHP10-3 cells. Cell proliferation activity was examined by CCK-8 colorimetric assays,  and the cell cycles of the stimulated cells were analyzed by flow cytometer. The expression of Cyclin D1 protein was observed by western-blot,  and the expression of cyclinD1 mRNA was observed by RT-PCR. Results   Compared with the control group,  the absorbance of the potassium iodate group,  the BPA group and the potassium iodate+BPA group was higher (P<0.05), the proportion of cells in the G0-G1 phase decreased and proportion of cells in the S phase increased significantly (P<0.05), the expression levels of Cyclin D1 protein and mRNA were up-regulated significantly(P<0.05). Conclusion   The potassium iodate and BPA can promote the proliferation of thyroid papillary carcinoma cells in a certain concentration range.
