Effect of Edaravone on autophagy in midbrain of Parkinson′s mice model

Abstract:

Objective   To investigate the effect of Edaravone on autophagy in midbrain of Parkinson′s disease (PD) mice. Methods   Male C57BL/6 mice were randomly divided into the model group, the Edaravone-treated group and the normal control group. The model group and the Edaravone-treated group were intraperitoneally injected with paraquat (10mg/kg) and maneb (30mg/kg) twice weekly for 6 consecutive weeks. Subsequent Edaravone-treated group were intraperitoneally injected with edaravone (3mg/kg) once a day for 2 consecutive weeks. At the same time the normal control group were intraperitoneally injected with the same dose saline. The behavior changes and the motor coordination were observed, and the expressed levels of LC3Ⅱ and beclin1 in the midbrain were detected by Western blot. Results   Administration of Edaravone significantly attenuated the impairment of the behavior performance, and decreased the expressions of LC3Ⅱ and beclin1 in the midbrain. Conclusion   The present findings suggest that Edaravone has a beneficial influence on parkinsonism by down regulating autophagy in the midbrain.
