Association of  killerimmunoglobulinlike receptors in the decidua and unexplained early recurrent spontaneous abortion

Abstract:

To study the relationship between killerimmunoglobulinlike receptors in the decidua and early unexplained recurrent spontaneous abortion(URSA). MethodsDecidual samples were obtained from 36 patients suffering from unexplained recurrent spontaneous abortion and 34 normal pregnant women. Genespecific PCR amplification (PCRSSP) was used to determine the individuam KIR genotypes  in women experiencing URSA and the controls. Reverse transcription polymerase chain reaction(RTPCR) was used to detect the individual KIR mRNA. KIR2DL1 mRNA and KIR2DL3 mRNA levels in the decidua from URSA and normal pregnancies were quantified by real time RTPCR. Results① Prevalence of activating KIR genes was higher in URSA  patients than in the controls. Among women experiencing URSA, the expressive rate of KIR2DS1 was 60.27% in patients and 41.18% in the controls(P<0.05). ② Transcripts of inhibitory KIRs mRNA were detected by RTPCR in individuals. The frequencies of inhibitory KIRs mRNA did not differ between women with URSA and the normal controls. No activating KIRs mRNA was detected. ③ The relative level of KIR2DL1 mRNA did not show significant differences between the two groups (699.37±259.78 vs 1?002.96±476.98, P<0.05), but the KIR2DL3 mRNA did (625.93±234.81 vs 1216.12±425.96, P>0.05). ConclusionsThe increased frequency of KIR2DS1 may be one of the reasons for URSA. A genetic imbalance between activating and inhibitory KIR genes might confer susceptibility to the occurrence of abortion.
