苦参碱对人结肠癌细胞株HT-29体外侵袭能力的抑制作用

Inhibitory effect and mechanism of matrine on the invasive ability of the colon cancer cell line HT-29 in vitro

摘要: 目的:探讨苦参碱对人结肠癌细胞株HT-29体外侵袭能力及乙酰肝素酶(heparanase, HPSE)基因表达的影响。方法:HT-29细胞体外培养,以0.125、0.25、0.5mg/ml不同浓度苦参碱进行预处理48h后,采用半定量RT-PCR、Western-blot检测各组培养细胞HPSE mRNA及HPSE蛋白表达的变化,采用细胞黏附实验检测细胞黏附能力的改变,Transwell小室检测侵袭及趋化能力的变化。结果:与对照组比较,各浓度苦参碱处理组HT-29细胞HPSE mRNA及HPSE蛋白的表达和细胞黏附侵袭能力均受到显著的抑制(P<0.01),其抑制效应与药物的浓度呈正相关(组间比较P<0.01)。结论:苦参碱能够显著抑制细胞的体外侵袭能力,其机制可能与下调HT-29细胞HPSE基因表达有关。

Abstract: Objective: To investigate the effect of matrine on the invasiveness and expression of heparanas-mRNA in the human colon cancer cell line HT-29. Methods: HT-29 cells were subjected to matrine of different concentrations. Fortyeight hours later, the expression of heparanase-mRNA was determined by semi-quantitative RTPCR, and the level of HPSE protein by western blot. The effect of matrine on HT29 cells was tested by cell Matrigel adhesion assay. The chemotactic migration and the invasive ability of HT-29 cells were determined by Matrigel invasion assay. Results: The expression of hepanase-mRNA and hepanase protein, adhesion, chemotactic migrations and invasiveness of matrine treated HT-29 cells significantly decreased compared with those of the controls(P<0.01). The inhibitory effect was dose-dependent(P<0.01). Conclusion: By down-regulating the expression of heparanase-mRNA and hepanase protein, matrine has a significant inhibitory effect on the adhesion and invasiveness of the human colon cancer cell line HT-29 in a dose-dependent manner within a certain range.
