重组骨形成发生蛋白-7促进种植体骨组织界面形成实验研究

Promoting osteogenesis by recombined human BMP-7 at the interface of oral implants

摘要: 目的:观察重组骨形成发生蛋白-7(BMP-7)对种植体骨组织界面形成的作用。方法:构建种植体植入动物模型,以正常侧作为对照,应用组织学方法观察种植体骨结合界面形态,组织化学方法测定碱性磷酸酶(ALP)活性,电子探针X射线能谱分析系统测定界面元素变化。结果:实验组在2周时,ALP活性较高,2至6周时迅速升高接近峰值,逐渐稳定直至12周。而对照组均匀升高,两组在各个时间段ALP活性差异都有统计学意义(P<0.05)。组织学观察发现,2周时种植体骨界面就有新生骨组织形成,6周时种植体界面已形成大量新生骨组织,种植体与机体组织之间只有极其薄的一层纤维膜,已达到骨结合标准。电子探针X射线能谱分析表明,种植体植入后6周内,钙元素与磷元素逐渐增加并达到峰值,以后趋于平稳。而对照组在各项检测中都较实验组迟缓。结论:BMP-7可以加速骨种植界面的愈合。

Abstract: Objective: To investigate the role of recombined human BMP-7 in the bone formation of implanted interface microregion. Methods: A histological microscope was used to observe the morphology, the histological chemistry method was used to determine the ALP activity and electronic probe X -ray microregion analysis was used to evaluate the element of the interface. Results: Two weeks after the implantation, an amount of new bone formation was observed through the histological microscope and at 6 weeks it nearly reached the bone integrated. The ALP activity was very high at two weeks after the implantation and almost reached a peak value at 6 weeks, and then it remained steady. The percentage of Ca and P increased rapidly and reached a peak at 6 weeks, and then it remained steady and persisted with the time. Conclusion: Recombination human BMP-7 may promote bone interface formation in oral implants.
