摘要: 目的 探究GPR56的Stachel peptide(P19)对米色脂肪组织棕色化的影响及其分子机制。 方法 利用qPCR和Western blotting方法检测GPR56在米色脂肪组织中的表达水平。用ELISA方法检测50 μmol/L P19激活小鼠米色脂肪组织中GPR56的Gq通路而导致的下游3-磷酸肌醇(inositol triphosphate, IP3)水平变化。随后,在诱导原代米色脂肪细胞棕色化分化过程中,通过qPCR方法检测棕色化标志基因和产热标志基因的表达水平,对P19、YM-254890(YM)和沉默Gpr56基因的表达对分化进程的影响进行综合分析。 结果 GPR56在米色脂肪组织中高表达。P19可以使棕色化标志基因(Tbx和Tmem26)和产热标志基因(Ppargc1a、Cox7a1和Cidea)表达水平升高,差异有统计学意义(P<0.05)。此外,当加入YM或沉默Gpr56基因的表达时,这些棕色化标志基因和产热标志基因的表达水平出现了反转。 结论 P19能够通过激活GPR56的Gq通路来促进米色脂肪组织的棕色化过程。
