血清瘦素、肿瘤坏死因子α、IL-6在高脂诱导肥胖大鼠胰岛素抵抗中的作用

摘要: 的:探讨高脂诱导肥胖模型大鼠血清瘦素、肿瘤坏死因子α(TNFα)、白细胞介素6(IL6)的水平及其在胰岛素抵抗中的作用。方法:大鼠高脂饮食8周后,以体重比对照组重30?g为标准,分出肥胖大鼠,继续饲养4周,空腹麻醉下,心脏取血,分离血清。葡萄糖氧化酶法测血清血糖;放射免疫法(RIA)测血清瘦素、胰岛素;酶联免疫法(ELISA)测血清TNFα、IL6水平。以空腹胰岛素/血糖(Insulin/Glucose,I/G)比值作为胰岛素敏感指标。结果:与对照组相比,肥胖大鼠空腹胰岛素、I/G比值,瘦素、IL6水平显著高于对照组(P均<0.05),血清TNFα水平无差别;肥胖大鼠I/G比值与血清瘦素、IL6均密切相关(r=0.772, P=0.036; r=0.898, P=0.015),与TNFα不相关(r=0.341, P=0.254)。结论:肥胖大鼠血清瘦素、IL6显著增高,参与了肥胖相关的胰岛素抵抗,血清TNFα未增高,与肥胖相关的胰岛素抵抗无关。

Abstract: To evaluate the role of serum leptin, TNFα and IL6 on insulin resistance in highfat diet induced obese rats. Methods: After fed with highfat diet for 8 weeks, the obese rats who were 30g heavier than controls were separated and fed for another 4 weeks. The blood samples were drawed by heart puncture. The serum levels of glucose were measured by glucose oxideperoxidase method, the levels of leptin and insulin by RIA, and the levels of TNFα, IL6 by ELISA. The ratio of insulin versus glucose(I/G) was used as an indicator for insulin resistance. Results: Compared with the controls, the serum levels of leptin, IL6 and I/G were higher in the obese rats, but the levels of TNFα were not different between two groups; The levels of leptin and IL6,but not TNFα, were closely related to the parameters evaluating insulin resistance in obese rats. Conclusions: The serum levels of leptin and IL6 are increased in highfat diet induced obese rats and contribute to insulin resistance. The levels of TNFα in obese rats are neither statistically increased nor related to insulin resistance.
