脂氧合酶抑制剂NDGA对HT29结肠癌细胞转移影响的研究

摘要: 12 3.宁津县中医院外科,山东 宁津 253400)目的:探讨脂氧合酶抑制剂去甲二氢愈创木酸(nordihydroguaiaretic acid,NDGA)在体外对结肠癌HT29细胞株转移的影响。方法:应用不同浓度的NDGA处理HT29结肠癌细胞,RTPCR检测血管内皮生长因子(vascular endothelial growth factor,VEGF),流式细胞仪检测CD44v6表达的变化;扫描电镜观察癌细胞表面微绒毛的变化。结果:经过NDGA作用后,CD44v6、VEGF随着药物浓度的增大,表达逐步下调。扫描电镜检测NDGA处理癌细胞48?h后,癌细胞微绒毛卷曲成小球状。结论:NDGA能够抑制HT29结肠癌细胞转移。

Abstract: To investigate the effect of the lipoxygenase inhibitor, nordihydroguaiaretic acid (NDGA) on metastasis of colon cancer cell line HT29 in vitro. Methods: Different concentrations of NDGA were used in the cancer cell. RTPCR was used to detect vascular endothelial growth factor(VEGF), flow cytometry to detect the expression of CD44v6, and scanning electron microscope to observe cells microvilli. Results: The expression of CD44v6 went down with the rise of concentration and so was VEGF detected with RTPCR. Through scanning electron microscope, it was found that cells microvilli curled to microball after NDGA had been used for 48 hours. Conclusion: NDGA can suppress the metastasis of colon cancer cellline HT29.
