海人酸致痫大鼠海马CA3区神经元线粒体损伤及caspase3的表达

摘要: 目的:观察海人酸(kanic acid,KA)诱导的癫痫持续状态(status epilepticus,SE)大鼠海马CA3区神经元线粒体超微结构的损伤及caspase3表达的变化。方法:用KA诱导大鼠SE 2?h,分别于SE终止后第3、6、24?h取脑,用电镜观察海马CA3区线粒体的超微结构,同时用免疫组化方法检测相同部位caspase3表达的变化。结果:SE终止后3?h可见到海马线粒体超微结构损伤,从肿胀到膜的完整性破裂。caspase3的表达在SE后6?h开始增加,于第24小时明显增高。结论:在实验性SE模型中,海马线粒体的超微结构在早期即有损伤,caspase3在线粒体损伤后数小时才出现表达,并于SE后24?h明显增高。

Abstract: [ABSTRACT]Objective: To observe the mitochondrial ultrastructure damage and caspase3 expression in the neurons of hippocampal CA3 subarea during kainic acid(KA) induced status epilepticus(SE). Methods: SE was induced in adult male Wistar rats with KA, and then 3, 6, and 24 hours later, the brain sections were made. The mitochondrial ultrastructure was observed with electron microscope, and the caspase3 expression was examined by immunohistochemical staining. Result: Mitochondrial ultrastructure damage could be seen, from swelling to the rupture of membrane 3 hours after SE, whereas caspase3 expression appeared 6 hours after SE and increased significantly at the 24hour point after SE. Conclusion: Mitochondrial ultrastructure is damaged in the early period of the experimental SE rats, and the caspase3 expression appeared a few hours later.
