亚低温对过氧化氢损伤后脑微血管内皮细胞NF-κB表达的影响

摘要:

目的   研究亚低温对过氧化氢损伤后脑微血管内皮细胞核转录因子(NF-κB)表达的影响。方法   分离并培养大鼠脑微血管内皮细胞,用过氧化氢损伤的方法建立脑出血后血肿周围炎症反应内皮细胞模型。将细胞分为实验组(ICH)和对照组(C)。ICH组分为过氧化氢损伤常温组(ICH+N)和过氧化氢损伤亚低温组(ICH+H),C组分为常温对照组(C+N)和亚低温对照组(C+H)。采用Western blot技术检测各组NF-κB的表达水平。结果   实验组NF-κB的表达水平均高于各自对照组(P<0.01), ICH+N组蛋白表达水平高于ICH+H组(P<0.01)。结论   亚低温可有效降低过氧化氢损伤的脑微血管内皮细胞NF-κB的表达,从而减轻血肿周围炎症反应,发挥其脑保护作用。

Abstract:

Objective   To investigate the influence of mild hypothermia on the expression of nuclear factor-kappa B(NF-κB) in cerebral microvascular endothelial cells injured by hydrogen peroxide. Methods   The cells of rat cerebral microvascular endothelial were separated and cultured, and the cell model of inflammatory response after cerebral hemorrhage was established by the method of hydrogen peroxide injury. Cells were divided into the experiment group(ICH) and the control group(C). Each group was divided into the normothermia  group(N ) and the mild hypothermia group(H). The expression of NF-κB was detected by western blotting after hydrogen peroxide intervention. Results   The expression of NF-κB obviously increased in the experiment group compared with the control group(P<0.01). While the level of  NF-κB was higher in the normothermia group than that in the mild hypothermia group(P<0.01).  Conclusion   Mild hypothermia may protect perihematoma tissues through relieving the inflammation by inhibiting NF-κB over-expression.
