己酮可可碱对大鼠胰腺纤维化的影响

摘要: 大鼠胰管内注射2%三硝基 苯磺酸(TNBS),制备大鼠胰腺纤维化模型。将大鼠12只随机分为PTX干预组、对照组。PTX干预组于术后第2天给予PTX 6?mg/(kg.d)腹腔注射 ,4周后收集两组胰腺标本。采用免疫组化技术检测治疗前后胰腺组织中α平滑肌肌动蛋白(αSMA)、血小板衍化生长因子(PDGF)、Ⅰ 型胶原(COLⅠ)、磷酸化细胞外信号调节激酶1(PERK1)的含量同时做组织胶原纤维染色,部分胰腺组织液氮冻存,以Western blot方法检测 αSMA的表达。结果:对照组发生胰腺纤维化, PTX干预组有轻度纤维化。PTX干预组αSMA、PDGF、COLⅠ的含量以及PERK1在基质细胞中的 表达均低于对照组(P<0.01)。胶原纤维染色显示,PTX治疗组胶原面积百分比明显低于对照组(P<0.01)。结论:PTX有抗大鼠胰腺纤维化的

Abstract: o observe the effects of pentoxifylline on pancreas fibrosis in rat. Methods: TNBS in a concentration of 2% was injected into the rats′pancreatic duct to set up the model of pancreatitis. Rats of interventional group were treated with PTX, and the expression of PDGF, αSMA, COL I and PERK1 were determined by using immunohistochemistry technique and HE and Masson staining in some samples 4 weeks later, meanwhile the expression of αSMA was detected by using Western blotting. Results: Pancreas fibrosis was observed in control group, whereas it was not significant in PTX treatment group. Compared with the control, PTX significantly reduced both the content of αSMA, PDGF, COL I and PERK1(P<0.01)and the pancreas fibrosis proportion(P<0.01). Conclusion: Pancreas fibrosis can be inhibited by PTX in rats.
