Effects of mycophenolate mofetil on neural function of experimental autoimmune encephalomyelitis in rats

Abstract: To explore the effects of mycophenolate mofetil on the improvement of neural function and brain and spine cord of rats with experimental autoimmune encephalomyelitis (EAE). MethodsWistar rats were induced by guinea pig spinal cord homogenate and complete freund adjuvant to establish EAE models, while the control rats were induced by saline and complete freund adjuvant. There were 7 control rats and 25 EAE rats which were successfully made and survived. The EAE rats were randomly divided into three groups: the high dose of mycophenolate mofetil treatment group(n=8), the low dose of mycophenolate mofetil treatment group(n=8), and the model group(n=9). Medicine was administered for 14 days and then the Duckers score and weight changes were assessed every day. After 14 days, pathological changes of brain and spine cord were observed by Hematoxylin and Eosin staining. ResultsThe achievement ratio of the EAE model was 72.1%. Compared with the control group, the Duckers score of the EAE rats increased with a decrease of weight(P<0.01), pathological changes manifested inflammatory cells infiltration around vessels with HE staining. Compared with the model group, after treatment with mycophenolate mofetil, the Duckers score of the EAE rats decreased with an increase of weight and pathological changes lessened, moreover, the therapeutic effect of high dose mycophenolate mofetil was superior to low dose mycophenolate mofetil(P<0.01). ConclusionMycophenolate mofetil can improve the symptoms of EAE rats and promotes the recovery of neural function in a dosedependent manner.
