Effect of DC-CIK pulsed with tumor lysate antigens of  SKOV3 on cytotoxic activity against SKOV3

Abstract:

Objective  To investigate the effect of dendritic cells(DC) pulsed with tumor lysate antigens of human ovarian cancer cell line SKOV3 co-cultured with cytokine induced killer(CIK) cells on cytotoxic activity against SKOV3. Methods  Blood isolated from 12 patients with ovarian cancer was separately induced to obtain CIK and DC. Unstimulated DC and DC pulsed with tumor lysate antigens of SKOV3 were individually co-cultured with CIK. The surface molecule expressions of DC in the DC group, Ag-DC group and  Ag-DC-CIK group, and the surface molecule expressions of CIK in the CIK group, DC-CIK group and Ag-DC-CIK group were measured by flow cytometry. The cytotoxic activities against SKOV3 of the CIK group, DC-CIK group and Ag-DC-CIK group were measured using lactate dehydrogenase(LDH) assay. Results  Compared with the DC group and the Ag-DC group, the Ag-DC-CIK group had a significant increase of mature phenotypes of DC(P<0.01); the mature phenotypes of CIK in the Ag-DC-CIK group also increased(P<0.01); the cytotoxic activity against SKOV3 cells of the Ag-DC-CIK group was much higher than that of the CIK group and the DC-CIK group(P<0.01). Conclusion  DC pulsed with tumor lysate antigens of SKOV3 co-cultured with CIK cells can promote the mutation of both DC and CIK and improve cytotoxic activity against SKOV3.
