Effect of fetal rat fibroblasts on wound healing of deep second degree scalds treated with dermabrasion

Abstract:

Objective  To investigate the effect of rat fibroblast transplant on wound healing. Methods  Forty eight experimental animals were inflicted with two deep second degree scalding wound with hot water(99℃, 10s) on the back of each rat. The burn wounds were dermabraded and divided into two groups: group A(n=48) was treated with transplanted Ffbs and dermabrasion, and group B(n=48) was treated with adults fibroblasts and dermabrasion. Samples were harvested on days 0,1,3,7,14 and 28 after burns. Changes of wound healing (T), fluorescence signs area(A), desensity(D=gray scale n/gray scale 0) and level of TGF-βmRNA were observed. Fetal rat skin tissue samples were harvested and digested with trypsin to isolate fibroblasts. The cultured cells were identified by vimentin with the immunohistochemical method and then labeled with CM-dil. The labeled fFBs were transplanted to burn wounds. Results  The fluorescence area began to expanded on 3 DPB(A=15.46±1.21)while its desensity began to decrease(D: 0.65±0.02), the fluorescence area reached its largest area(A=16.69±1.00)with its much lower desensity (D=0.31±0.01) on day 14, and slender fluorescence signals was still determined on day 28(D=0.09±0.01). TGF-β1 mRNA in group A increased in response to burns and decreased from day 1. In group B, however, TGF-β1 mRNA reached a climax on day 7 and wasmaintained at a relatively high level duringstudy period. The wound healing period was 8.13±0.99 days in group A and 10.25±1.04 days in group B. Conclusion  Fetal fibroblast can survive and proliferate in xenogenic wound tissue. TGF-β1 mRNA expression in the wound tissue with transplanted fibroblasts is depressed.
