RNA interference inhibits VEGF expression in retinoblastoma cells: an experimental study

Abstract:

To explore the possibility of VEGF as a therapeutic target for retinoblastoma, we employed RNA interference to inhibit VEGF expression in retinoblastoma cells. MethodsSiRNA oligo of VEGF was synthesized and transfected into retinoblastoma cells. VEGF mRNA expression and protein content were respectively determined by reverse transcription polymerase chain reaction(RTPCR) and Western blot. Retinoblastoma cell proliferation was detected by MTT reduction assay. ResultsVEGF mRNA expression and protein content were significantly decreased by VEGFtargeted siRNA. MTT reduction assay showed that the inhibition ratio was 40%. ConclusionVEGFtargeted siRNA can effectively inhibit expression of VEGF in retinoblastoma cells.
