Osteopontin promotes the invasiveness of human ovarian
carcinoma cell line SKOV3: an experimental study

Abstract: To investigate the expression of Osteopontin (OPN) and explore its effect on the invasiveness of human ovarian carcinoma cell line SKOV3. MethodsHuman ovarian carcinoma SKOV3 cells were transfected with plasmid pcDNA 3.1(-)/OPN and cells transfected with a mock plasmid served as controls. RTPCR and Westernblot were used to determine the expressions of OPN mRNA and proteins. Cell invasiveness was measured by the matrigel invasion assay. ResultsExpressions of OPN mRNA and protein were significantly increased (P<0.01) after transfection. Migrating numbers of SKOV3 cells transfected with pcDNA 3.1(-)/OPN were also significantly increased (P<0.05) as compared with the control group. ConclusionOPN play an important role in the metabasis of ovarian cancer.
