Effect of striatal extracts on inducing the NSC differentiate into dopaminergic neurons: an experimental study

Abstract: ObjectiveTo culture human fetal neural stem cells in vitro, and then induce them to differentiate into dopaminergic neurons. Methods Human fetal neural stem cells derived from the hippocampus were purified and induced by human fetal striatal extracts. The induced differentiational rate of dopaminergic neurons was detected by immunocytoflurescence and the expression of TH- mRNA was determined by RT-PCR. Results The rate of TH+ cells of the experimental group was higher than that of the control group, and the expression of TH-mRNA was not obvious in the control group but was strong in the induced group. There were significant differences between the two groups. Conclusion Human fetal striatal extracts can effectively induce human fetal NSCs to differentiate into dopaminergic neurons in vitro.
