姜黄素对糖尿病大鼠肾脏结构和功能的影响

摘要: 目的观察姜黄素对糖尿病大鼠肾脏结构和功能的影响。方法用链脲佐菌素(STZ)诱导制备糖尿病大鼠模型,并随机分为糖尿病组和姜黄素治疗组,另设一正常对照组。治疗组给予姜黄素200?mg/(kg·d)灌胃,为期8周。然后进行肾脏形态学观察和血尿素氮(BUN)、血肌酐(Scr)、内生肌酐清除率(Ccr)、尿白蛋白排泄率(UAER)等的检测。结果糖尿病组大鼠Ccr、UAER、尿白蛋白/肌酐、肾小球平均截面积(MGA)、肾小球平均体积(MGV)均显著高于正常对照组(P<0.01或P<0.05)。光镜和电镜下观察显示,糖尿病组大鼠较正常对照组大鼠肾小球明显增大,细胞数目增多,基底膜节段性增厚,部分足细胞足突融合。姜黄素治疗组上述异常均明显减轻。三组大鼠间血BUN和 Scr两两比较无明显差异(P>0.05)。结论姜黄素可明显改善糖尿病大鼠肾脏病理改变,减轻肾脏肥大和高滤过状态,降低尿白蛋白,对糖尿病大鼠有肾脏保护作用。

Abstract: To explore the effect of curcumin on the structure and function of the kidney in diabetic rats. MethodsDiabetes was induced by a single intraperitoneal injection of streptozoticin(STZ) to male Wistar rats at a dose of 60?mg/kg body weight and they were randomly divided into two groups: the diabetes group (group D) and the diabetes treated with curcumin group(group DC). A normal control group (group N) was also designed. Curcumin was given at a dose of 200?mg·kg-1·day-1, the other two groups were given the same quantity of CarboxylmethylCellulose(CMC). After a treatment of 8 weeks, renal morphology and blood and urinary biochemistry were evaluated. ResultsLevels of clearance of creatinine(Ccr), urinary albumin excretion rate(UAER), albumincreatinine ratio(Alb/Cr), mean glomerular area(MGA) and mean glomerular volume(MGV) were all significantly increased in group D as compared with group N(P<0.01 or P<0.05) . Large glomeruli,membranes with a thickened glomerular basement, and foot processes confluence into podocytes were found by light microscopy and electron microscopy. All these abnormalities were markedly ameliorated after a curcumin treatment. ConclusionCurcumin is effective in improving renal pathological damage, ameriorating hypertrophy and hyperfiltration, and decreasing the urinary albumin excretion rate(UAER)in diabetic Wistar rats.
