转录因子T-bet/GATA-3诱导哮喘模型中α-SMA和Ⅰ型胶原高表达的实验研究

High expression of α-SMA and type Ⅰ collagen induced by transcription factor T-bet/GATA-3 ratio in asthma models

摘要: 目的观察大鼠肺转录因子T细胞表达的T盒(T-bet)/GATA连接蛋白-3 (GATA3)对肺支气管α-平滑肌动蛋白(α-SMA)和Ⅰ型胶原的调解作用,探讨其比率与哮喘早期气道重塑的关系。方法将大鼠随机分为OVA组即哮喘模型组、生理盐水组、空白对照组。采用实时定量-聚合酶链反应法检测大鼠肺组织中T-bet mRNA和GATA-3 mRNA表达,免疫印迹法检测大鼠肺组织中T-bet和GATA-3蛋白表达;免疫组化法测定大鼠肺组织中α-SMA表达水平,酶联免疫吸附法测定成纤维细胞(LF)中Ⅰ型胶原表达强度;比较T-bet/GATA-3与α-SMA和Ⅰ型胶原的关系。结果OVA组大鼠肺组织中T-bet mRNA和蛋白表达强度均较生理盐水组和空白对照组显著减弱;而OVA组GATA3 mRNA和蛋白表达强度较其余两组显著增强;T-bet/GATA-3比率较其余两组显著减弱;OVA组肺组织α-SMA和LF中Ⅰ型胶原含量较其余两组显著增强;T-bet/GATA-3比率与α-SMA和Ⅰ型胶原均成负相关。结论 T-bet/GATA-3比率与哮喘早期气道重塑的指标α-平滑肌动蛋白(α-SMA)和Ⅰ型胶原等呈线性负相关,T-bet/GATA-3失衡可能是哮喘早期气道重塑的重要原因之一。

Abstract: ObjectiveTo study the relationship between transcription factor T-bet/GATA -3 and expression of α-smooth muscle actin (α-SMA) and type Ⅰ collagen in asthma. MethodsThe levels of T-bet and GATA-3 in rat lungs were determined by quantitative real-time PCR and western blotting respectively. α-SMA and type Ⅰ collagen levels in lung fibroblasts were determined by immunohistochemistry and ELISA respectively. ResultsThe expression of T-bet mRNA and protein in the OVA group was significantly attenuated compared with that in the saline group and the blank group, while the expression of GATA-3 in the OVA group was significantly increased and the T-bet/GATA-3 ratio in the OVA group was significantly decreased. The α-SMA and type Ⅰ collagen levels in the OVA group were significantly increased. The T-bet/GATA-3 ratio had a negative correlation with α-SMA and type Ⅰ collagen. There was no significant difference between the saline group and the blank group. ConclusionThe T-bet/GATA3 ratio has a negative relation to α-SMA and type Ⅰ collagen, and T-bet/GATA3 imbalance plays a crucial role in asthma models.
