羟基红花黄色素A对四氯化碳急性肝损伤的保护作用

Protective effect of hydroxysafflor yellow A on carbon tetrachlorideinduced hepatotoxicity

摘要:

目的探讨羟基红花黄色素A(HSYA)对四氯化碳(CCl4)急性肝损伤的保护作用。方法将60只昆明种小鼠随机分为正常对照组和CCl4损伤组、HSYA保护组(2、4、8、16?mg/kg组),每组各10只。CCl4腹腔注射建立小鼠急性肝损伤模型。观察肝组织病理学改变;测定血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)的活性,肝组织匀浆谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、丙二醛(MDA)、肿瘤坏死因子α(TNFα)的含量及肝细胞色素P450 2E1(Cytochrome P450 2E1,CYP2E1)蛋白的表达。结果与CCl4损伤组比较,HSYA保护组肝组织病理改变明显减轻;血清ALT、AST活性明显降低(P<0.05); 肝组织匀浆中GSH、SOD含量明显升高(P<0.05);MDA、TNFα含量明显降低(P<0.05);CYP2E1蛋白表达量增加。综合各指标表明,以8?mg/kg保护组效果最佳。结论HSYA对CCl4急性肝损伤有明显保护作用,其机制可能与抑制脂质过氧化,清除自由基,降低TNFα水平有关。

Abstract:

To investigate the protective effect of Hydroxysafflor Yellow A(HSYA) against Carbon tetrachloride (CCl4)induced hepatotoxicity and its mechanism.  MethodsA total of 60 mice were randomly and averagely divided into 6 groups: the blank control group, the CCl4 group, four dosages of HSYA groups (2, 4, 8, 16?mg/kg). 10% CCl4 was used to induce acute liver injury in all groups expect for the control group. All animals were sacrificed after 24h from CCl4 intraperitoneal injection. Liver injury was assessed by elevations of serum alanine aminotransferase (ALT) and aspartate aminotranferase (AST) activities and histopathology. Antioxidative activity was measured by the formation of malondialdehyde (MDA).  Superoxide anion scavenging activity was evaluated by contents of glutathione (GSH) and superoxide dismutase (SOD). Radioimmunoassay (RIA) was used to measure formation of tumor necrosis factorα(TNFα). Expression level of cytochrome P450 2E1 (CYP2E1) protein was measured by western blot analysis.  ResultsPretreatment of HSYA was shown to possess a significantly protective effect by significantly lowering the serum AST and ALT (P<0.05). This hepatoprotective action was confirmed by histological observations. In addition, pretreatment of HSYA significantly prevented the elevation of MDA formation and the depletion of GSH and SOD contents in the liver of CCl4injected mice (P<0.05). The groups pretreated with HSYA significantly developed lower TNFα levels compared with nonHSYA pretreated CCl4injected mice (P<0.05). Expression level of CYP2E1 in the HSYA pretreated groups was higher than that in the nonHSYA pretreated CCl4injected group, but was lower compared with the blankcontrol group.  ConclusionHSYA has a potent hepatoprotective effect on CCl4induced acute liver injury in mice through antioxidant activity and reducing formation of TNFα.
