Silence of MHC gene has an effect on the cytotoxicity of LAK cells by small interfering RNA duplexes in mice

Abstract: ObjectiveTo investigate the relationship between major histocompatibility complex (MHC) expression of lymphokine activated killer (LAK) cells and the cytotoxicity in vitro. MethodsH-2Kd-targeting small interfering RNA (siRNA ) was applied to spleen lymphocytes of BABL/C mice. Flow cytometry (FCM) was used to determine the expression of H-2Kd on the transfected and control cells. The cytotoxicity for H22 and K562 cell lines of the transfected cells was determined in vitro by the LDH release assay. ResultssiRNA treatment for H-2Kd gene significantly decreased the expression level of the targeted protein, pure trans-messenger and non-silencing siRNA did not inhibit the H-2Kd expression at the tested concentrations. The cytotoxicity for H22 and K562 cells was significantly decreased. ConclusionsiRNA-reduced down-expression of H-2Kd may have some relation to LAK cells cytotoxicity for neoplasm cell lines.
