Grape seed proanthocyanidin extract combined with atorvastatin in prevention of atherosclerosis in rabbits

Abstract: ObjectiveTo investigate the anti-atherogenic effects of atorvastatin combined with grape seed proanthocyanidin extract (GSPE) on atherosclerosis, and!to explore its possible mechanism. MethodsThe total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol（HDL-c）, and low density lipoprotein cholesterol（LDL-c） were determined by electronic chemiluminescene, and the content of malondialdehyde （MDA）was determined by TBA. At the end of the twelfth week, the aorta and the heart of the rabbits were excised for morphological observation by an optical microscope and an electron microscope and by an immunohistochemical method. ResultsIn the GSPE combined with atorvastatin group, the serum MDA content was significantly decreased(P＜0.01）, and the serum lipid profile was more significantly decreased than that in the GSPE group（P＜0.05）. The atherosclerosis was obviously relieved by GSPE combined with atorvastatin. The atherosclerosis was better inhibited in the GSPE combined with atorvastatin group than in the atorvastatin group. ConclusionsAtorvastatin combined with GSPE could obviously alleviate the development of arteriosclerosis, and it is prior to GSPE or atorvastatin only.
