Effect of sampling positions in nucleic acid test on the test result: a report from 47 confirmed COVID-19 cases in Jinan City

Abstract: Objective To investigate the effect of sampling positions in nucleic acid test for COVID-19 on test results. Methods Clinical specimens collected from 47 confirmed cases of COVID-19 in Jinan City during Jan. 22 and Feb. 11, 2020 were tested with real-time fluorescence quantitative RT-PCR to analyze the effect of different sampling positions on the test results. Statistical analysis was conducted with Fisher’s exact test. Results The 47 patients included 23 males and 24 females, age ranging from 11 months to 71 years, average 35 years. The positive rate of nasopharyngeal swabs and sputum samples was 91.11%(41/45)and 92.31%(13/14), respectively. There was no statistical difference between the results of the two types of samples(Fishers exact test, bilateral P>0.999). However, the Ct value was lower in sputum samples than in nasopharyngeal swabs. Conclusion The nasopharyngeal swabs can meet the requirements for screening and diagnosis of COVID-19, and deep-coughed sputum samples can be used to re-test the “false negative” nasopharyngeal swabs.
