Effects of sleeve gastrectomy on lung injury in type 2 diabetic rats

Abstract: Objective To investigate the effects of sleeve gastrectomy on lung injury in type 2 diabetic rats and to explore the mechanism. Methods Type 2 diabetic models were induced by intraperitoneally injecting streptozotocin(STZ)to Wistar rats. The diabetic rats were randomly assigned to the sleeve gastrectomy group(SG group, n=10)and Sham group(n=10). The body weight, food intake, fasting blood glucose(FBG)and glucose tolerance were measured postoperatively. In postoperative week 16, the rats were sacrificed. The lung tissues were collected and the histomorphology was observed with HE staining. The expressions of inflammation factors interleukin-6(IL-6), tumor necrosis factor α(TNF-α)and oxidative stress indicator nitrotyrosine(NT)were determined with Western blotting and immunohistochemistry. The expressions of nitric oxide(NO)and malondialdehyde(MDA), as well as the activity of superoxide dismutase(SOD)were determined with biochemical testing. Results Only 1 rat in the SG group died after surgery. Compared with the sham group, SG group had better glucose control(FBG: F=100.70, P<0.01, AUCOGTT: F=88.15, P<0.01), mitigated thickness of interalveolar septum, reduced hemorrhage, lower expressions of IL-6, TNF-α, NT, NO and MDA(P<0.01), but higher activity of SOD(P<0.01). Conclusion Sleeve gastrectomy can relieve the lung injury in type 2 diabetic rats, possibly by inhibiting the expressions of inflammation factors and improving oxidative stress in lungs.
