Inhibitory effect of simvastatin on expression of Cytokine TNFα,
IL6 and uPA/uPAR secreted by HUVECs

Abstract:

To explore the effect of simvastatin on expressions of TNFα, IL6, uPA/uPAR mRNA and protein in HUVECs treatment with LPS. MethodsCultivated HUVECs were divided into two groups: the 12?h group and the 24?h group. Then they were subdivided into four groups: the control group, the LPS（100?ng/mL）group, the simvastatin(5?μg/mL) group, and the LPS and simvastatin group. Expressions of TNFα, IL6, uPA/uPAR mRNA and protein were determined using realtime quantitative PCR and ELISA. ResultsCompared with the control groups, expressions of TNFα, IL6, uPA/uPAR mRNA and IL6 in the supernatant were significantly increased in the LPS groups (P<0.05). Compared with the LPS groups, expressions of TNFα, IL6, uPA/uPAR mRNA and IL6 in the supernatant were statistically decreased in the LPS and simvastatin groups (P<0.05). ConclusionSimvastatin can inhibit expressions of TNFα, IL6 and uPA/uPAR secreted by HUVECs.
