Effects of IL17 neutralization antibody on polymorphnuclear of asthma in mice

Abstract: To investigate the effects of IL17 neutralization antibody on polymorphnuclear (PMN) of asthma mice. MethodsA total of 40 Kunming mice were randomly divided into four groups: the control group (group A), the asthma group (group B), the IL17 neutralization antibody group (group C), and the Dex group (group D).Mice were sensitized and challenged with ovalbumin(OVA) to establish asthma models, and groups C and D were injected with the IL17 neutralization antibody and Dex respectively. Peripheral blood PMN was isolated and purified to observe the apoptosis percentage. The total cell number and differentiation cell number were counted in the bronchoalveolar lavage fluid (BALF) . The pathological characters of lung tissues were observed using HE staining. Myeloperoxidase (MPO) expression was detected by immunohistochemical technique. ResultsPercentage of the PMN apoptosis in group C was significantly higher than that in group B, while the number of Neu in BALF and MPO level in lung tissues were significantly decreased. Compared with group B, the percentage of PMN apoptosis and number of Neu in BALF in group C had no significant differences, while levels of MPO were significantly increased. ConclusionIL17 neutralization antibody may inhibit asthmatic airway inflammation caused by PMN.
