Interaction between polymorphism in the ADH1B gene and hypertriglyceridemia in premature coronary artery disease

Abstract: To assess the interaction between the ADH1B*1/2 polymorphism and hypertriglyceridemia (HTG) in premature coronary artery disease (pCAD). MethodsA hospitalbased casecontrol study was conducted. Newly diagnosed CAD patients were recruited as the subjects. The 167 CAD patients diagnosed before or at 55 years old for males and 65 years old for females were assigned as the pCAD, i.e. the case group; the other 235 lateonset CAD patients served as the control group. TG over 2.26?mmol/L was defined as hypertriglyceridemia. Polymerase chain reactionrestriction fragment length polymorphism was used to detect the polymorphisms. A multivariate logistic regression model was performed to adjust the potential confounders and estimate odds ratios (ORs). Synergy index (S), relative excess risk due to interaction (RERI) and attributable proportion due to interaction (AP) were the measures to assess the interaction as departure from additivity. ResultsThere was a positive interaction between the ADH1B*1/2 polymorphism and HTG in pCAD. S was 1.72, RERI was 1.78 and AP was 33.77%. After adjusting for sex, smoking, alcohol drinking, BMI, total cholesterol and diastolic blood pressure by multiple logistic regression, there was also a positive interaction. S was 2.96, RERI was 3.01 and AP was 54.29%. ConclusionThe interaction between the ADH1B*1/2 polymorphism and HTG had a positive effect on pCAD in this studied population.
