Protective effect of gypenosides-containing serum against oxidation neurotoxicity induced by glutamate

Abstract: To investigate the neuroprotective effects of gypenosides (GP) containing serum on glutamate (Glu) induced oxidation toxicity to cortical neurons. Methods Sera containing GP was collected by ways of seropharmacology, and the primary cortical neurons isolated from Wistar rats E1415d embryos were studied. The viability of cells was detected by the MTT method, the cellular death pathway was analyzed by Ho33342 and PI doublelabeled method, the contents of SOD and GSH were determined by biochemistry method, and Cytosolic [Ca2+] was determined by flow cytometry (FCM). ResultsCompared with Glu, GPcontaining serum could increase the cell survival rate and block the decrease of SOD and GSH and the increase of cytosolic[Ca2+]. Conclusion GP-containing serum can prevent the neurons from damage of glutamate oxidative neurotoxicity and the 400mg/kg/d GP-containing serum has the strongest antagonistic action.
