肝细胞生长因子及其受体在口腔鳞癌和颈淋巴结中的表达

Expressions of HGF and its receptor in oral squamous cell carcinoma and neck lymph nodes

摘要: 目的通过检测肝细胞生长因子(HGF)及其受体c—Met在口腔鳞癌中的表达水平,探讨HGF/c-Met系统对口腔鳞癌侵袭转移的影响。 方法对50例确诊的原发、复发口腔鳞癌病例标本,采用免疫组织化学(SABC)法检测口腔鳞癌组织和颈部淋巴结中HGF和cMet的表达情况。将收集的病例按病理类型及临床资料分组,并比较各组间HGF和c-Met的表达差异。 结果口腔鳞癌组织及有转移灶的淋巴结中,癌细胞HGF及c-Met蛋白的阳性表达率为100%,颈部淋巴结中HGF的阳性率为60%,淋巴细胞内均无c-Met蛋白的表达。癌细胞c-Met蛋白的表达水平和颈淋巴结内淋巴细胞HGF表达阳性与口腔鳞癌的局部淋巴结转移高度相关(P=0.0000),癌细胞中HGF的高水平表达与肿瘤的侵袭(P=0.0020)、复发相关(P=0.0139)。 结论口腔鳞癌中HGF和c-Met蛋白表达升高、颈部淋巴结中淋巴细胞HGF表达阳性可能促进肿瘤的侵袭、复发、局部淋巴结转移,提示患者预后不良。

Abstract: To explore the effect of HGF/cMet system on invasive and metastasis potential of oral squamous cell carcinoma (OSCC). MethodsFifty biopsy samples were collected from primary and relapse OSCC patients. Immunohistochemical staining was used to determine the expression of hepatocyte growth factor (HGF) and c-Met protein in OSCC tissues and neck nodes. Samples were divided into groups based on clinicopathological features and then the expression intensity was analyzed between groups. Results The expression rate of HGF and c-Met in carcinoma cells was 100%. The HGF expression rate in neck nodes was 60%. However, c-Met was not expressed in lymphocytes of neck nodes. High expression level of HGF in carcinoma cells and positive expression of c-Met in lymphocytes of neck lymph nodes significantly correlated with lymph nodes metastasis (P=0.0000), and high expression level of HGF had trends to invade (P=0.0020) and relapse(P=0.0139). Conclusion Overexpression of HGF and c-Met in OSCC and positive expression of c-Met in lymphocytes of neck nodes may induce invasion, recurrence and metastasis, predicting a poor prognosis.
