血清外泌体miR-20b-5p对非小细胞肺癌的诊断价值

Diagnostic value of serum exosomal miR-20b-5p for non-small cell lung cancer

摘要: 目的 探讨血清外泌体miR-20b-5p对非小细胞肺癌(NSCLC)患者的诊断价值。 方法 收集144例NSCLC患者(NSCLC组)血清和138例健康对照人群(健康对照组)血清。分离并鉴定血清外泌体。提取血清外泌体miRNA并通过RT-qPCR比较NSCLC组和健康对照组之间miR-20b-5p的表达水平。利用受试者工作特征曲线(ROC)评估miR-20b-5p的诊断价值。 结果 纯化的外泌体具有已报道的外泌体的典型特征。NSCLC组外泌体miR-20b-5p的表达水平显著高于健康对照组,差异有统计学意义(P<0.001)。ROC曲线下面积(AUC)为0.849(95% CI:0.802~0.889,敏感度为73.61%,特异度为70.29%)。 结论 血清外泌体miR-20b-5p在NSCLC中高表达,且具有较高的临床诊断价值,可以作为辅助NSCLC诊断的新型生物标志物。

Abstract: Objective To explore the diagnostic value of serum exosomal miR-20b-5p for non-small cell lung cancer(NSCLC). Methods Serum samples of 144 NSCLC patients(NSCLC group)and 138 healthy controls(healthy group)were collected. Exosomes in the serum were extracted and characterized, and miRNA was abstracted. The expression of exosomal miR-20b-5p in the NSCLC group and healthy control group were compared with RT-qPCR. The diagnostic value of exosomal miR-20b-5p was evaluated with receiver operating characteristic(ROC)curve. Results Purified exosomes had typical features. The expression of miR-20b-5p was significantly higher in the NSCLC group than in the healthy control group (P<0.001). The area under the ROC curve(AUC)was 0.849(95%CI:0.802~0.889). The diagnostic sensitivity was 73.61%, and the specificity was 70.29%. Conclusion Serum exosomal miR-20b-5p is highly expressed in NSCLC with highly clinical diagnostic value. It can serve as a new biomarker to assist the diagnosis of NSCLC.
