下调α-catulin基因的表达对舌鳞癌细胞株Tscca侵袭及迁移能力影响的体外研究

Down-regulation of α-catulin inhibits the invasion and migration of human tongue squamous cancer cell line Tscca in vitro

摘要: 目的 探讨α-catulin基因下调对人舌鳞癌细胞株Tscca侵袭、迁移能力的影响。 方法 本实验设为3个组:α-catulin-siRNA组、空质粒转染组和未转染质粒的空白对照组。构建α-catulin沉默质粒转染的人舌鳞癌细胞株Tscca,运用Western blotting检测α-catulin蛋白表达, 采用Transwell及划痕实验检测细胞的侵袭、迁移能力。 结果 Western blotting结果显示,α-catulin-siRNA转染组Tscca细胞α-catulin蛋白表达量减少;Transwell及划痕实验结果显示,α-catulin-siRNA转染组细胞侵袭、迁移能力降低。 结论 下调α-catulin基因的表达可以有效降低人舌鳞癌细胞株Tscca的侵袭、迁移能力,α-catulin有望成为舌癌基因治疗的一个潜在靶点。

Abstract: Objective To explore the effect of α-catulin down-regulation on the invasion and migration of tongue squamous cancer cell line Tscca. Methods Cells were divided into α-catulin-siRNA transfection group, empty plasmid transfection group, and blank group. The α-catulin down-expressing plasmid was constructed and transfected into human tongue squamous cancer cell line Tscca. The protein expression of α-catulin was tested by Western blotting. The in vitro invasion and migration abilities of Tscca cells were measured by Transwell assays and Wound Healing. Results The protein expression of α-catulin reduced remarkably in α-catulin-siRNA trasfection group. Wound-healing and Transwell assays showed that the invasion and migration of Tscca cells were suppressed in α-catulin-siRNA transfection group. Conclusion SiRNA-mediated silencing of the α-catulin gene can significantly inhibit cell invasion and migration in human squamous cell carcinoma line Tscca, indicating that α-catulin might be used as an underlying target for molecular therapy of tongue cancer.
